Abstract:
Glycosylation plays pivotal roles in both eukaryotic and prokaryotic organisms. Employing new-to-nature chemistry to decipher biological functions of glycan, as well as to fulfil glycan-based therapeutic need, remains a major challenge. We will present the development of a liposome-assisted bioorthogonal reporter strategy (LABOR) to achieve selective visualization and detection of eukaryotic glycans both in vitro and in vivo. Azidosugars were encapsulated within ligand-targeted liposomes, and selectively delivered into target cells in a receptor-mediated manner. The delivered unnatural monosaccharide derivatives were then metabolically incorporated into cell surface glycans. Cases in detection of tissue-specific and organ-specific sialoglycans using LABOR will be discussed. Also, we deciphered the membrane-to-membrane transport process of lipopolysaccharide (LPS), a Gram-negative cellular glycolipid, by expanding the genetic code of amino acids on the lipopolysaccharide transport proteins (Lpt proteins). Our results support a model in which LPS are pushed one after the other in a PEZ dispenser-like manner across a protein bridge.
